Dosing
Acne Vulgaris ABSORICA: 0.5 to 1 mg/kg/day in two divided doses for 15 to 20 weeks (up to 2 mg/kg/day for severe disease with scarring or trunk involvement); ABSORICA LD: 0.4 to 0.8 mg/kg/day in two divided doses for 15 to 20 weeks (up to 1.6 mg/kg/day for severe disease).
Acne Vulgaris Apply a pea-sized amount to the face once daily; wash face gently with mild soap, rinse with warm water, and pat dry before applying. Avoid eyes, mouth, mucous membranes, and areas of broken skin.
Contraindications
- Pregnancy
- Hypersensitivity to isotretinoin, Vitamin A (given chemical similarity), or any component of the product
- Hypersensitivity (e.g., anaphylaxis) to clindamycin, benzoyl peroxide, any components of the formulation, or lincomycin
- History of regional enteritis, ulcerative colitis, or antibiotic-associated colitis
Adverse Reactions

Pharmacology
Isotretinoin is a retinoid that inhibits sebaceous gland function and keratinization; clinical improvement in nodular acne is associated with a reduction in sebum secretion, reflecting reduced sebaceous gland size and inhibition of sebaceous gland differentiation.
Clindamycin is a lincosamide antibacterial that binds to the 50S ribosomal subunit of susceptible bacteria and suppresses protein synthesis by interfering with peptidyl transfer; benzoyl peroxide is an oxidizing agent with bactericidal and keratolytic effects, though its precise mechanism is unknown.
