Dosing
Rheumatoid Arthritis, Arthritis, Psoriatic, Ankylosing spondylitis 40 mg every other week SC; some RA patients not receiving MTX may benefit from 40 mg every week or 80 mg every other week.
Crohn Disease, Ulcerative Colitis 160 mg on Day 1, 80 mg on Day 15, then 40 mg every other week SC starting Day 29.
Psoriasis vulgaris, Panuveitis 80 mg initial dose, then 40 mg every other week SC starting 1 week after initial dose.
Multiple Sclerosis 300 mg IV infusion over one hour every four weeks.
Crohn Disease 300 mg IV infusion over one hour every four weeks; discontinue if no therapeutic benefit by 12 weeks of induction therapy or if concomitant corticosteroids cannot be tapered within six months of starting therapy.
Contraindications

- Progressive multifocal leukoencephalopathy (PML), current or historical
- Previous hypersensitivity reaction to TYSABRI, ranging from urticaria to anaphylaxis

Serious Serious infections (pneumonia, septic arthritis, cellulitis, diverticulitis, pyelonephritis), malignancies, acute liver failure, hepatitis B reactivation, neurologic reactions, hematological reactions, heart failure, autoimmunity
Postmarketing Diverticulitis, large bowel perforations, pancreatitis, liver failure, autoimmune hepatitis, sarcoidosis, Merkel Cell Carcinoma, demyelinating disorders, cerebrovascular accident, interstitial lung disease, pulmonary embolism, Stevens Johnson Syndrome, cutaneous vasculitis, erythema multiforme, new or worsening psoriasis, alopecia, systemic vasculitis, deep vein thrombosis
Most common (>=10%) Headache, fatigue (MS and CD); arthralgia, urinary tract infection, lower respiratory tract infection, gastroenteritis, vaginitis, depression, pain in extremity, abdominal discomfort, diarrhea, rash (MS); upper respiratory tract infection, nausea (CD).
Serious Progressive multifocal leukoencephalopathy (PML), herpes infections, hepatotoxicity, hypersensitivity and antibody formation, immunosuppression and infections, hematological abnormalities.
Postmarketing Hemolytic anemia, thrombocytopenia (including immune thrombocytopenic purpura).
Pharmacology
Adalimumab-adbm is a TNF-alpha antagonist that binds specifically to TNF-alpha and blocks its interaction with the p55 and p75 cell surface TNF receptors, modulating inflammatory responses and reducing concentrations of acute phase reactants such as CRP and ESR.
Alpha4-integrin antagonist; recombinant humanized IgG4 monoclonal antibody that binds to the alpha4-subunit of alpha4beta1 and alpha4beta7 integrins on leukocytes, blocking their adhesion to VCAM-1 and MAdCAM-1 and inhibiting transmigration into inflamed parenchymal tissue in multiple sclerosis and Crohn's disease.
