Pharmacology
Enfuvirtide is an HIV-1 fusion inhibitor that binds to the first heptad-repeat (HR1) domain of the gp41 subunit of the viral envelope glycoprotein, preventing the conformational changes required for fusion of viral and cellular membranes and thereby blocking HIV-1 entry into CD4+ cells.
Lenacapavir is a multistage, selective HIV-1 capsid inhibitor that directly binds to capsid protein (p24) subunits in hexamers, inhibiting multiple essential steps of the viral lifecycle including capsid-mediated nuclear uptake of proviral DNA, virus assembly and release, and capsid core formation.
