Dosing
Psoriasis vulgaris, Arthritis, Psoriatic 6 mg orally once daily, with or without food; do not crush, cut, or chew tablets; not recommended in severe hepatic impairment (Child-Pugh C).
Psoriasis vulgaris 160 mg SC at Week 0, then 80 mg at Weeks 2, 4, 6, 8, 10, 12, followed by 80 mg every 4 weeks.
Arthritis, Psoriatic 160 mg SC at Week 0, followed by 80 mg every 4 weeks.
Ankylosing spondylitis, Non-Radiographic Axial Spondyloarthritis 160 mg SC at Week 0, followed by 80 mg every 4 weeks.
Contraindications
  • History of hypersensitivity reaction to deucravacitinib or any excipient in SOTYKTU
  • Previous serious hypersensitivity reaction to ixekizumab or to any of the excipients
Adverse Reactions
Most common (>=1%) Upper respiratory infections, blood creatine phosphokinase increased, herpes simplex, mouth ulcers, folliculitis, acne
Serious Pneumonia, COVID-19, malignancies including breast cancer, hepatocellular carcinoma, and lymphoma
Most common (>=1%) Injection site reactions, upper respiratory tract infections, nausea, tinea infections.
Serious Serious infections, hypersensitivity reactions including anaphylaxis, inflammatory bowel disease.
Pharmacology
TYK2 inhibitor; deucravacitinib binds the regulatory domain of TYK2, allosterically inhibiting receptor-mediated TYK2 activation and downstream STAT signaling via the JAK-STAT pathway.
Humanized IgG4 monoclonal antibody that selectively binds to interleukin-17A (IL-17A) and inhibits its interaction with the IL-17 receptor.
