Dosing
Psoriasis vulgaris, Dermatitis, Atopic Apply a thin layer to affected areas once daily; wash hands after application unless VTAMA cream is for treatment of the hands; not for oral, ophthalmic, or intravaginal use.
Psoriasis vulgaris Apply ZORYVE cream, 0.3%, to affected areas once daily in adult and pediatric patients 6 years of age and older.
Dermatitis, Atopic Apply ZORYVE cream, 0.15%, once daily for patients 6 years of age and older; apply ZORYVE cream, 0.05%, once daily for pediatric patients 2 to 5 years of age.
Contraindications

  • Moderate to severe liver impairment (Child-Pugh B or C)
Adverse Reactions
Most common (>=1%) - Plaque Psoriasis Folliculitis, nasopharyngitis, contact dermatitis, headache, pruritus, influenza
Most common (>=1%) - Atopic Dermatitis Upper respiratory tract infection, folliculitis, lower respiratory tract infection, headache, asthma, vomiting, ear infection, pain in extremity, abdominal pain

Pharmacology
Tapinarof is an aryl hydrocarbon receptor (AhR) agonist; the specific mechanisms by which VTAMA cream exerts its therapeutic actions in plaque psoriasis and atopic dermatitis are unknown.
PDE4 inhibitor; roflumilast and its active metabolite (roflumilast N-oxide) inhibit PDE4, a major cyclic AMP-metabolizing enzyme, leading to accumulation of intracellular cyclic AMP.
