Dosing
Diabetes Mellitus Inject subcutaneously in the abdominal region, buttocks, thigh, or upper arm twice daily within 15 minutes before meal initiation (or after meal initiation for type 2 patients); individualize dose based on metabolic needs, blood glucose monitoring results, and glycemic control goal.
Diabetes Mellitus, Type 1 Approximately one-third of total daily insulin requirements SC once daily at the same time each day; use concomitantly with short-acting premeal insulin.
Diabetes Mellitus, Type 2 0.2 units/kg or up to 10 units SC once daily at the same time each day for insulin-naive patients; individualize based on metabolic needs and glycemic goals.

- Episodes of hypoglycemia
- Hypersensitivity to Novolog Mix 70/30 or one of its excipients
- Episodes of hypoglycemia
- Hypersensitivity to insulin glargine products or any excipient in Semglee
Adverse Reactions
Most common (>=5%) Hypoglycemia, headache, influenza-like symptoms, dyspepsia, back pain, diarrhea, pharyngitis, rhinitis, skeletal pain, upper respiratory tract infection, neuropathy, abdominal pain
Serious Severe hypoglycemia requiring assistance or parenteral glucose/glucagon, anaphylaxis, lipodystrophy, localized cutaneous amyloidosis
Postmarketing Localized cutaneous amyloidosis at injection site, hyperglycemia with repeated injections into areas of amyloidosis, hypoglycemia with sudden change to unaffected injection site, medication errors due to accidental substitution

Pharmacology
Biphasic insulin analog consisting of 70% insulin aspart protamine (prolonged absorption) and 30% soluble insulin aspart (rapid absorption); lowers blood glucose by stimulating peripheral glucose uptake in skeletal muscle and fat and by inhibiting hepatic glucose production.
Insulin glargine-yfgn is a long-acting human insulin analog that lowers blood glucose by stimulating peripheral glucose uptake in skeletal muscle and fat and inhibiting hepatic glucose production; it also inhibits lipolysis and proteolysis and enhances protein synthesis.
