Dosing
Lymphoma, T-Cell, Cutaneous Initial dose 300 mg/m2/day as a single oral daily dose with a meal; may be reduced to 200 or 100 mg/m2/day or temporarily suspended for toxicity; may be escalated to 400 mg/m2/day after 8 weeks if no tumor response and 300 mg/m2/day is well tolerated.
Lymphoma, T-Cell, Cutaneous Start with once every other day; increase weekly to once, twice, three, then four times daily as tolerated; apply a generous coating to cover lesions, avoiding normal surrounding skin and mucosal surfaces; do not use occlusive dressings.

Adverse Reactions

Serious Hyperlipidemia, pancreatitis, hepatotoxicity, cholestasis, hepatic failure, hypothyroidism, neutropenia, cataracts, hypoglycemia in patients with diabetes mellitus, photosensitivity
Most common (>=10%) Rash, pruritus, skin disorder, pain, infection, headache, contact dermatitis, edema, hyperlipemia

Bexarotene selectively binds and activates retinoid X receptor subtypes (RXRalpha, RXRbeta, RXRgamma), which function as transcription factors regulating genes that control cellular differentiation and proliferation; the exact mechanism of action in CTCL is unknown.
