Dosing
Migraine Disorders 50 mg or 100 mg orally as needed; a second dose may be taken at least 2 hours after the initial dose; maximum 200 mg per 24-hour period. Dose reductions apply with moderate or weak CYP3A4 inhibitors, BCRP and/or P-gp only inhibitors, and in patients with severe hepatic or severe renal impairment (CLcr 15-29 mL/min); avoid in end-stage renal disease.
Migraine Disorders One packet (50 mg) dissolved in 1 to 2 ounces (30 to 60 mL) of water, taken orally as a single dose on an empty stomach for best effectiveness; use the lowest effective dose for the shortest duration consistent with individual patient treatment goals.
Contraindications
- Concomitant use of strong CYP3A4 inhibitors
- History of serious hypersensitivity to ubrogepant or any component of UBRELVY (reactions have included anaphylaxis, dyspnea, and facial or throat edema)
- Known hypersensitivity (e.g., anaphylactic reactions and serious skin reactions) to diclofenac or any components of the drug product
- History of asthma, urticaria, or other allergic-type reactions after taking aspirin or other NSAIDs
- In the setting of coronary artery bypass graft (CABG) surgery

Pharmacology
Ubrogepant is a calcitonin gene-related peptide (CGRP) receptor antagonist; by blocking CGRP receptor signaling it inhibits a pathway implicated in migraine pathophysiology.
NSAID that inhibits cyclooxygenase (COX-1 and COX-2), reducing prostaglandin synthesis in peripheral tissues; analgesic, anti-inflammatory, and antipyretic properties are attributed to this decrease in prostaglandins, which sensitize afferent nerves and mediate inflammation.
