Dosing
Diabetic Retinopathy, Uveitis Single 0.19 mg (190 mcg) fluocinolone acetonide intravitreal implant injected under aseptic conditions via a preloaded single-use applicator; optimal placement is inferior to the optic disc and posterior to the equator, with needle entry 4 mm inferotemporal from the limbus.
Macular retinal edema, Uveitis One 0.7 mg intravitreal implant injected into the vitreous cavity under controlled aseptic conditions; each applicator is single-use and treats one eye only.
Contraindications
  • Active or suspected ocular or periocular infections, including active epithelial herpes simplex keratitis (dendritic keratitis), vaccinia, varicella, mycobacterial infections, and fungal diseases
  • Glaucoma with cup to disc ratio greater than 0.8
  • Known hypersensitivity to any components of ILUVIEN
  • Active or suspected ocular or periocular infections, including viral diseases of the cornea and conjunctiva (active epithelial herpes simplex keratitis, vaccinia, varicella), mycobacterial infections, and fungal diseases
  • Glaucoma with cup to disc ratios greater than 0.8
  • Torn or ruptured posterior lens capsule due to risk of migration into the anterior chamber
  • Known hypersensitivity to any components of this product

Pharmacology
Fluocinolone acetonide is a synthetic corticosteroid that inhibits inflammatory responses by blocking phospholipase A2 via lipocortins, suppressing biosynthesis of prostaglandins and leukotrienes and reducing edema, leukocyte migration, capillary proliferation, and fibroblast proliferation.
Dexamethasone is a corticosteroid that suppresses inflammation by inhibiting multiple inflammatory cytokines, resulting in decreased edema, fibrin deposition, capillary leakage, and migration of inflammatory cells.
